Method of 4A molecular sieve desiccant
Conduct integrity testing on the glove system of the sterile isolator, as well as the experimental and transfer chambers.
Using the same batch of 4A molecular sieve desiccant for continuous sterilization and dehumidification for 3 times, verify the bactericidal effect of hydrogen peroxide on the isolator by detecting the concentration, distribution status, and sterilization effect of hydrogen peroxide gas, as well as the internal environment microorganisms (settling bacteria, planktonic bacteria, surface microorganisms) of the isolator.
By weighing the 4A molecular sieve before and after use and observing the fogging phenomenon in the cabin, the effective number of times the 4A molecular sieve achieves sterilization and dehumidification effect determined.
Result
A total of 8 glove systems tested for integrity, and the pressure drop difference of the gloves within 60 seconds was less than 60 Pa.
The integrity test results of the experimental chamber and transfer chamber showed an hourly volume leakage rate of 0.04%.
And the temperature fluctuation in each measurement environment was less than 0.5 ℃.
The hydrogen peroxide indicator card shows that the hydrogen peroxide in the cabin has reached the sterilization concentration and evenly distributed.
According to biological indicator testing, hydrogen peroxide can kill more than 106 thermophilic Bacillus subtilis.
The results of microbial detection in the internal environment of the isolator are as follows: sedimentation bacteria have 0 colony forming units (CFU) every 4 hours, planktonic bacteria have 0 CFU/m3, and surface microorganisms have 0 CFU/plate.
The weight of the same batch of 4A molecular sieve desiccant before use was 3.000 5 kg, and the weights after the first, second, and third uses were 3.185 0, 3.340 5, and 3.451 5 kg, respectively. When the same batch of desiccant used continuously for the third time, fogging occurred in the isolator compartment.
Conclusion
The same batch of 4A molecular sieve desiccant can be used continuously for 3 times and achieve the expected sterilization and dehumidification effect.
